Glucosaminyl (N-acetyl) transferase 3 (GCNT3) is a key enzyme responsible for the synthesis of core 2 O-glycans, vital for mucin production and glycan branching on glycoproteins GCNT3 is primarily expressed in mucin-secreting epithelial cells of the gastrointestinal, respiratory, and reproductive tracts, where it regulates epithelial barrier integrity, mucus production, and protection against microbial invasion It has a critical role in modulating inflammatory responses, with altered GCNT3 activity implicated in diseases like colitis, Crohn’s disease, and chronic inflammation of mucosal surfaces GCNT3 also functions as a tumor suppressor, with reduced expression linked to cancers such as colorectal, pancreatic, and gastric cancers where disrupted glycosylation promotes tumor growth, invasion, and metastasis Emerging studies suggest GCNT3 regulates immune responses by maintaining epithelial glycan patterns essential for immune defense pathways Its role in glycan biosynthesis highlights its importance in epithelial protection, mucosal immunity, and tumor suppression pathways
