Descripción
GABBR2 is the partner subunit of the GABA-B receptor, forming a functional heterodimer with GABBR1. While GABBR1 is responsible for ligand binding, GABBR2 facilitates G protein activation and downstream signaling. GABBR2 couples to Gi/o proteins, leading to the inhibition of adenylate cyclase, reduced cAMP levels, and the opening of potassium channels while inhibiting calcium channels. This signaling reduces neuronal excitability and neurotransmitter release, mediating the inhibitory effects of GABA in the central nervous system. GABBR2 is highly expressed in brain regions involved in motor control, pain, and cognition, where it regulates synaptic plasticity and long-term depression (LTD). Dysregulation of GABBR2 has been implicated in disorders such as epilepsy, schizophrenia, depression, and drug addiction. GABA-B receptor modulators targeting GABBR2 are being explored for their therapeutic potential in neurological diseases, pain management, and mood disorders. The functional partnership of GABBR1 and GABBR2 highlights their combined role in maintaining inhibitory signaling and synaptic homeostasis in the brain.
