Descripción

GALR3 is a G protein-coupled receptor that primarily couples to Gi/o proteins, mediating the inhibitory effects of galanin by reducing intracellular cAMP levels and modulating ion channels. GALR3 is expressed in brain regions such as the hypothalamus, hippocampus, and amygdala, where it regulates mood, feeding, and reward processing. GALR3 activation has been implicated in the regulation of depression and anxiety, where it exerts an inhibitory effect on neuronal excitability and neurotransmitter release. In pain pathways, GALR3 contributes to the modulation of nociceptive signaling, though its role appears less defined compared to GALR1 and GALR2. GALR3 is also expressed in peripheral tissues, where it may influence cardiovascular function and immune responses. In cancer, GALR3 has been associated with tumor progression, particularly in breast and prostate cancers, where galanin signaling promotes cell survival and proliferation. Dysregulation of GALR3 contributes to mood disorders, neuropathic pain, and tumor growth. Its role in neuronal inhibition and emotional regulation makes GALR3 a promising target for treating mood disorders, pain, and cancer progression.

The neuropeptide galanin modulates a variety of physiologic processes including cognition/memory, sensory/pain processing, hormone secretion, and feeding behavior. The human galanin receptor Types are G protein-coupled receptor Types that functionally couple to their intracellular effector Type through distinct signaling pathways. GALR3 is found in many tissues and may be expressed as 1.4-, 2.4-, and 5-kb transcripts.
