Descripción
FZD8 is a member of the Frizzled receptor family that mediates Wnt signaling, predominantly through the canonical β-catenin-dependent pathway. It is expressed in various tissues, including the brain, heart, and bone, where it regulates cell proliferation, differentiation, and tissue homeostasis. FZD8 plays a significant role in bone formation and osteoblast differentiation, making it essential for skeletal development and regeneration. In cancer, FZD8 is overexpressed in lung, breast, and prostate cancers, where it promotes cell proliferation, survival, and therapy resistance through enhanced Wnt/β-catenin signaling. FZD8 also contributes to angiogenesis and metastasis by regulating endothelial cell function and tumor vascularization. Its role in brain development includes regulating neural differentiation and synaptic plasticity. FZD8 activation triggers downstream pathways like PI3K/Akt and MAPK, driving cellular responses critical for growth and repair. Dysregulation of FZD8 is associated with developmental defects, cancer progression, and bone disorders.
