Background
DOCK10 is a GEF for Rac1 and Cdc42, involved in regulating actin cytoskeletal dynamics, cell migration, and lymphocyte polarization. It is highly expressed in hematopoietic cells and epithelial tissues, promoting B-cell activation, dendritic cell migration, and immune surveillance. DOCK10 mediates filopodia and lamellipodia formation, enhancing membrane protrusion during cell migration. It is implicated in immune responses, particularly in B and T cells, where it supports polarization and chemotaxis. Dysregulation is associated with immune dysfunction and cancer metastasis through enhanced cell motility. Knockout models show defects in lymphocyte migration and immune cell activation.
