Background

Damage-specific DNA binding protein 2 (DDB2) is a substrate recognition protein that works in concert with DDB1 within the UV-DDB complex, a subset of the CUL4-DDB1 E3 ubiquitin ligase system. DDB2 plays an essential role in nucleotide excision repair (NER) by recognizing UV-induced DNA damage, such as cyclobutane pyrimidine dimers (CPDs) and 6-4 photoproducts, and recruiting repair factors to the damage site. Its high affinity for damaged DNA and ability to induce chromatin remodeling are key to facilitating repair processes. DDB2 also mediates the ubiquitination and degradation of histones and other chromatin-associated proteins, promoting access to damaged DNA. Mutations or dysregulation of DDB2 have been linked to xeroderma pigmentosum group E (XP-E), a condition characterized by extreme sensitivity to UV light and a high predisposition to skin cancer. Beyond DNA repair, DDB2 is implicated in cell cycle regulation and apoptosis, highlighting its multifunctional role in maintaining genomic integrity.
