Descripción
DENN domain containing 2D (DENND2D) is a guanine nucleotide exchange factor (GEF) within the DENN protein family, specifically targeting Rab GTPases, including Rab9. DENND2D plays a pivotal role in retrograde transport by activating Rab9 to facilitate vesicle movement between late endosomes and the trans-Golgi network. This transport is critical for recycling membrane proteins, maintaining lysosomal enzyme trafficking, and regulating receptor degradation. DENND2D is involved in cellular processes such as autophagy, endosomal sorting, and signaling pathways linked to cellular stress and growth. Its expression pattern suggests a broad functional relevance, particularly in tissues with active vesicular transport systems. Mutations or dysregulation of DENND2D have been associated with lysosomal storage disorders and other conditions resulting from impaired vesicle trafficking, underlining its importance in cellular health and homeostasis.
