(CMKLR1), also known as ChemR23, is a G-protein-coupled receptor (GPCR) primarily activated by chemerin, a protein involved in immune responses and adipose tissue regulation. CMKLR1 is expressed in immune cells, such as macrophages, dendritic cells, and natural killer (NK) cells, as well as in adipocytes and smooth muscle cells. Its signaling plays a crucial role in inflammation and immune cell migration, especially during tissue injury or infection. CMKLR1 activation promotes macrophage recruitment to inflamed tissues, aiding in immune responses and wound healing. It also influences adipogenesis and metabolic regulation, contributing to energy balance and insulin sensitivity. CMKLR1 is implicated in cardiovascular diseases, where it regulates vascular inflammation and smooth muscle contraction, affecting blood pressure and vessel remodeling. Dysregulation of CMKLR1 signaling has been linked to metabolic disorders such as obesity, type 2 diabetes, and chronic inflammation. Moreover, CMKLR1 is being investigated as a target in autoimmune diseases and cancers, where its modulation could impact tumor microenvironments and immune evasion.
