C-C Chemokine Receptor Type 8 (CCR8) is a G-protein-coupled receptor expressed predominantly on T-helper 2 (Th2) cells, regulatory T cells (Tregs), and subsets of monocytes and dendritic cells. CCR8 interacts primarily with its ligand CCL1, promoting the recruitment and migration of immune cells to inflammatory sites and tissues, particularly in the context of allergic responses and chronic inflammation. CCR8 plays a central role in Th2-mediated immune responses, contributing to diseases such as asthma, atopic dermatitis, and other allergic disorders. In addition, CCR8 is highly expressed on tumor-infiltrating Tregs in the tumor microenvironment, where it fosters immune suppression and allows tumor cells to evade host immunity. Its role in tumor progression and metastasis has made CCR8 an attractive target for cancer immunotherapy. Antibody-based therapies targeting CCR8 are under development to deplete Tregs specifically within tumors, restoring anti-tumor immune responses. CCR8 is also implicated in HIV infection, as it can serve as a co-receptor for the virus, facilitating viral entry into immune cells. Further understanding of CCR8 signaling and its interaction with CCL1 will help develop targeted therapies for allergic diseases, autoimmune conditions, and cancer, making it a versatile and critical player in immune regulation.

CCR8 encodes a member of the beta chemokine receptor family, which is predicted to be a seven transmembrane protein similar to G protein-coupled receptors. Chemokines and their receptors are important for the migration of various cell types into the inflammatory sites. This receptor protein preferentially expresses in the thymus. I-309, thymus activation-regulated cytokine (TARC) and macrophage inflammatory protein-1 beta (MIP-1 beta) have been identified as ligands of this receptor. Studies of this receptor and its ligands suggested its role in regulation of monocyte chemotaxis and thymic cell apoptosis. More specifically, this receptor may contribute to the proper positioning of activated T cells within the antigenic challenge sites and specialized areas of lymphoid tissues. CCR8 is located at the chemokine receptor gene cluster region.
