The adrenergic receptor beta-2 (ADRB2) is another G-protein-coupled receptor widely distributed in smooth muscle tissues, including the lungs, blood vessels, and uterus. ADRB2 activation by catecholamines induces smooth muscle relaxation through cAMP-mediated pathways, leading to bronchodilation, vasodilation, and uterine relaxation. In the respiratory system, ADRB2 is critical for airway relaxation, making it a primary target for bronchodilators used in asthma and chronic obstructive pulmonary disease (COPD) management. Additionally, ADRB2 plays a role in glycogenolysis in the liver and skeletal muscles, promoting energy mobilization during stress or exercise. Genetic variants in ADRB2, such as Gly16Arg and Gln27Glu, influence receptor sensitivity and have been associated with differential responses to beta-agonists and susceptibility to asthma. Overactivation of ADRB2 can lead to desensitization, reducing therapeutic efficacy over time. Its involvement in multiple physiological processes underscores ADRB2's importance in both metabolic and respiratory regulation.
