BAG cochaperone 5 (BAG5) is a member of the BAG family of cochaperones that interacts with heat shock proteins, particularly HSP70, to regulate protein folding, degradation, and cellular stress responses BAG5 plays a crucial role in managing the cellular proteostasis network, especially under stress conditions It has been implicated in modulating apoptosis, where it acts as a survival factor by influencing cell death pathways BAG5 regulates protein turnover by promoting the degradation of misfolded proteins and supporting cellular adaptation to oxidative stress Its expression has been linked to neurodegenerative diseases, where impaired protein quality control leads to toxic protein aggregates BAG5 is also involved in regulating muscle cell function and has been shown to play a role in protecting cells from atrophy and degeneration in muscle tissues Studies indicate that BAG5 is associated with cancer biology, where it can prevent apoptosis in tumor cells, contributing to resistance to chemotherapy and promoting tumor growth

BAG5 is a member of the BAG1-related protein family. BAG1 is an anti-apoptotic protein that functions through interactions with a variety of cell apoptosis and growth related proteins including BCL-2, Raf-protein kinase, steroid hormone receptors, growth factor receptors and members of the heat shock protein 70 kDa family. This protein contains a BAG domain near the C-terminus, which could bind and inhibit the chaperone activity of Hsc70/Hsp70. Three transcript variants encoding two different isoforms have been found for this gene.
