Description
BAG proteins compete with Hip for binding to the Hsc70/Hsp70 ATPase domain and promote substrate release. All the BAG proteins have an approximately 45-amino acid BAG domain near the C terminus but differ markedly in their N-terminal regions. The predicted BAG2 protein contains 211 amino acids. The BAG domains of BAG1, BAG2, and BAG3 interact specifically with the Hsc70 ATPase domain in vitro and in mammalian cells. All 3 proteins bind with high affinity to the ATPase domain of Hsc70 and inhibit its chaperone activity in a Hip-repressible manner. [provided by RefSeq].

BAG cochaperone 2 (BAG2) is a protein that functions as a cochaperone for heat shock proteins (HSPs), particularly HSP70, to regulate protein folding and degradation BAG2 plays a key role in controlling the cellular stress response by modulating protein homeostasis under stress conditions It interacts with misfolded proteins and assists in their proper folding or directs them for proteasomal degradation through the ubiquitin-proteasome pathway BAG2 also has roles in the regulation of apoptosis, where it can influence cell survival by interacting with pro-apoptotic factors BAG2 is implicated in various diseases, including cancer, where its expression can promote tumor progression by stabilizing key oncogenic proteins and inhibiting cell death pathways Additionally, BAG2 has been linked to neurodegenerative diseases such as Parkinson's disease, where its role in protein quality control is crucial for preventing the accumulation of toxic protein aggregates Its ability to modulate protein turnover and cell survival pathways highlights BAG2 as an important player in maintaining cellular integrity and a potential target for therapeutic intervention
