Description

ALOX5AP is a protein which, with 5-lipoxygenase, is required for leukotriene synthesis. Leukotrienes are arachidonic acid metabolites which have been implicated in various types of inflammatory responses, including asthma, arthritis and psoriasis. This protein localizes to the plasma membrane. Inhibitors of its function impede translocation of 5-lipoxygenase from the cytoplasm to the cell membrane and inhibit 5-lipoxygenase activation.

Descripción

Arachidonate 5-Lipoxygenase Activating Protein (ALOX5AP) is a crucial regulatory protein that facilitates the activity of 5-lipoxygenase (5-LOX), an enzyme involved in the biosynthesis of leukotrienes. Leukotrienes are lipid mediators derived from arachidonic acid that play significant roles in inflammatory and allergic responses, including bronchoconstriction, vascular permeability, and recruitment of immune cells. ALOX5AP acts as an essential cofactor, enabling 5-LOX to translocate to the nuclear membrane and interact with its substrate, leading to the production of leukotrienes. Dysregulation of ALOX5AP has been implicated in various inflammatory diseases, including asthma, atherosclerosis, and rheumatoid arthritis. Genetic variants in the ALOX5AP gene are associated with an increased risk of cardiovascular disease and stroke, making it a potential biomarker and therapeutic target. Inhibitors targeting ALOX5AP or its interaction with 5-LOX are being explored as treatments to mitigate leukotriene-mediated inflammatory conditions.
