Description

Ubiquitin-binding protein that interacts with the BRCA1-BARD1 heterodimer, and regulates its activity. Specifically binds 'Lys-6'-linked polyubiquitin chains. Interaction with autoubiquitinated BRCA1, leads to inhibit the E3 ubiquitin-protein ligase activity of the BRCA1-BARD1 heterodimer. Component of a complex required to couple deglycosylation and proteasome-mediated degradation of misfolded proteins in the endoplasmic reticulum that are retrotranslocated in the cytosol.

UBX domain protein 1 (UBXN1) is a ubiquitin regulatory protein containing a UBX domain, which enables its interaction with p97/VCP, a key ATPase involved in protein degradation and cellular quality control UBXN1 functions as a cofactor for p97, participating in the extraction and degradation of misfolded proteins during endoplasmic reticulum-associated degradation (ERAD) It regulates protein homeostasis by linking p97 to the ubiquitin-proteasome system and autophagy pathways UBXN1 is also involved in DNA damage repair, where it regulates protein turnover at replication forks and facilitates chromatin remodeling Dysregulation of UBXN1 impairs protein clearance, contributing to neurodegenerative diseases such as Alzheimer’s and Parkinson’s, where protein aggregates accumulate in neurons Emerging studies also connect UBXN1 to cancer biology, as it influences protein turnover in tumor cells and regulates apoptosis Its role in maintaining proteostasis and cellular stress responses underscores its importance in cellular quality control mechanisms
