Descripción
TMED4, a key p24 family member, participates in the regulation of vesicle-mediated trafficking in the early secretory pathway, facilitating cargo transport between the ER, ER-Golgi intermediate compartment (ERGIC), and Golgi apparatus. TMED4 interacts with coat proteins (COPII and COPI) to enable vesicle budding, cargo sorting, and retrieval, ensuring protein quality control and proper trafficking dynamics. It also contributes to maintaining Golgi organization, regulating secretory pathway efficiency, and supporting protein maturation processes. TMED4 is widely expressed across various tissues, particularly in actively secreting cells, where it supports cellular homeostasis and organelle function. Dysregulation of TMED4 is associated with neurodegeneration, metabolic disorders, and cancers, where altered trafficking pathways lead to protein mislocalization and cellular stress. Knockdown studies reveal disrupted vesicle formation, increased ER stress, and impaired protein transport, underscoring its role in coordinating intracellular trafficking, protein stability, and cellular function.
