ROBO3 (Roundabout Guidance Receptor 3) is a unique member of the Roundabout receptor family with critical roles in axon guidance and neural development. Unlike other ROBO proteins, ROBO3 facilitates the crossing of axons at the midline of the central nervous system by antagonizing Slit-mediated repulsion, a process essential for the development of bilateral neural circuits. ROBO3 expression is tightly regulated during neural development, and mutations in the ROBO3 gene have been linked to horizontal gaze palsy with progressive scoliosis, a rare congenital disorder affecting motor control and spinal development. This receptor’s ability to modulate axon guidance makes it indispensable for proper neural wiring and functional brain organization.
