Description
The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. Together with SUFU, prevents nuclear import of GLI1, and thereby inhibits GLI1 transcription factor activity. Regulates GLI1 in differentiating chondrocytes. Likewise, regulates GLI3 proteolytic processing and modulates GLI2 and GLI3 transcription factor activity. Plays a role in autophagic vacuole assembly, and mediates defense against pathogens, such as S.aureus, by promoting their capture by autophagosomes that then merge with lysosomes.

RAB23, member RAS oncogene family (RAB23) is a small GTPase that regulates the trafficking of intracellular vesicles and plays a role in the movement of molecules between the endosomal system and other cellular compartments, including the Golgi apparatus and plasma membrane RAB23 has been shown to participate in the process of autophagy, where it helps in the sorting and degradation of cellular materials via the lysosomal pathway, ensuring that damaged or unwanted components are properly processed and removed from the cell This function links RAB23 to cellular stress responses, maintaining cellular integrity and preventing the accumulation of damaged proteins and organelles Additionally, RAB23 is involved in regulating the function of the sonic hedgehog signaling pathway, which is critical for development and the control of cellular proliferation RAB23’s dysfunction is linked to various genetic disorders, including craniofacial malformations, where defects in its trafficking function disrupt proper cellular signaling and tissue development Furthermore, RAB23 is involved in cancer biology, where aberrant vesicular trafficking contributes to uncontrolled cell growth, tumor progression, and metastasis
