Description

Methyllysine-binding protein, component of the MOF histone acetyltransferase protein complex. Not required for maintaining the global histone H4 'Lys-16' acetylation(H4K16ac) levels or locus specific histone acetylation, but instead works downstream in transcriptional regulation of MOF target genes(By similarity). As part of the NSL complex it may be involved in acetylation of nucleosomal histone H4 on several lysine residues. Contributes to methyllysine-dependent p53/TP53 stabilization and up-regulation after DNA damage.

Descripción

PHF20 is a chromatin-binding protein that recognizes methylated histones, particularly H3K4me3 and H4K20me1, to regulate transcriptional activation, DNA repair, and cellular proliferation. It functions as a transcriptional co-activator by recruiting epigenetic modifiers and transcription machinery to target loci, ensuring precise gene expression during development and cellular responses. PHF20 is a key regulator of p53 stability and function, linking chromatin dynamics to tumor suppression and DNA damage responses. It is expressed in proliferative tissues and plays roles in stem cell maintenance, cell cycle regulation, and DNA repair. Dysregulation of PHF20 has been implicated in cancers, where its loss of function leads to genomic instability and uncontrolled proliferation. Knockout studies demonstrate defects in p53 signaling, impaired DNA repair, and reduced cellular proliferation, highlighting its essential role in chromatin regulation, tumor suppression, and transcriptional control.
