JADE1 is a key chromatin-associated protein involved in histone acetylation and transcriptional regulation as part of the HBO1 histone acetyltransferase complex. Through its PHD finger domain, JADE1 recognizes acetylated histone H4 and facilitates H3/H4 acetylation, regulating chromatin accessibility during transcription, DNA replication, and repair. JADE1 is highly expressed in tissues undergoing rapid cell division, such as embryonic and epithelial tissues, where it controls cell cycle progression and differentiation. It is also involved in the Wnt/β-catenin signaling pathway, where it promotes the stabilization of β-catenin, influencing gene expression programs critical for development and tissue regeneration. Dysregulation of JADE1 is associated with cancer progression, fibrosis, and developmental disorders due to altered chromatin modification and transcriptional regulation. Knockdown studies demonstrate defects in histone acetylation, impaired cell cycle control, and disrupted transcriptional activation, highlighting its essential role in maintaining chromatin structure, epigenetic regulation, and cellular homeostasis.
