Description

Adapter protein which associates with tyrosine-phosphorylated growth factor receptors, such as KDR and PDGFRB, or their cellular substrates. Maintains low levels of EIF2S1 phosphorylation by promoting its dephosphorylation by PP1. Plays a role in the DNA damage response, not in the detection of the damage by ATM/ATR, but for efficient activation of downstream effectors, such as that of CHEK2. Plays a role in ELK1-dependent transcriptional activation in response to activated Ras signaling. Modulates the activation of EIF2AK2/PKR by dsRNA. May play a role in cell adhesion and migration through interaction with ephrin receptors.

Descripción

NCK1 is a cytoplasmic adaptor protein containing three SH3 domains and one SH2 domain, which allows it to mediate interactions between receptor tyrosine kinases (RTKs) and downstream effectors in signaling pathways. NCK1 is involved in actin cytoskeleton reorganization, cell motility, and immune synapse formation by linking signaling proteins such as WASP, WAVE, and PAK1. It plays key roles in processes like cell migration, proliferation, and T-cell activation. NCK1 is highly expressed in various tissues and regulates growth factor receptor signaling, including pathways like MAPK/ERK and PI3K. Dysregulation of NCK1 is associated with cancer progression, metastasis, and immune system dysfunction. Knockout studies show defects in actin dynamics, impaired cell movement, and reduced T-cell activation, highlighting its importance in cytoskeletal signaling and cellular communication.
