DOCK3, or MOCA, is a neuron-specific GEF for Rac1 that regulates neurite outgrowth, axonal growth, and synaptic plasticity in the CNS. It mediates actin rearrangement, promoting axonal guidance and dendritic remodeling. Highly expressed in neurons, DOCK3 maintains synapse connectivity and supports neuroprotection. Dysregulation contributes to neurodegenerative diseases like Alzheimer’s, where it influences tau phosphorylation and cell survival pathways such as ERK/Akt activation. DOCK3 deficiency causes motor and cognitive impairments in mouse models, highlighting its role in neuronal development, CNS recovery, and axonal regeneration.
