Description
Beta-amyloid peptides are lipophilic metal chelators with metal-reducing activity. Bind transient metals such as copper, zinc and iron. In vitro, can reduce Cu(2+) and Fe(3+) to Cu(+) and Fe(2+), respectively. Beta-amyloid 42 is a more effective reductant than beta-amyloid 40. Beta-amyloid peptides bind to lipoproteins and apolipoproteins E and J in the CSF and to HDL particles in plasma, inhibiting metal-catalyzed oxidation of lipoproteins. Beta-APP42 may activate mononuclear phagocytes in the brain and elicit inflammatory responses. Promotes both tau aggregation and TPK II-mediated phosphorylation. Interaction with overexpressed HADH2 leads to oxidative stress and neurotoxicity. Also binds GPC1 in lipid rafts. The gamma-CTF peptides as well as the caspase-cleaved peptides, including C31, are potent enhancers of neuronal apoptosis.

Descripción
APP is a transmembrane glycoprotein widely expressed in the brain and peripheral tissues that plays a critical role in neuronal development, synaptic plasticity, and cell adhesion APP is proteolytically processed by enzymes such as α-secretase, β-secretase, and γ-secretase, leading to the production of various fragments, including the beta-amyloid (Aβ) peptide Abnormal processing of APP and the accumulation of Aβ peptides form amyloid plaques, which are a hallmark of Alzheimer’s disease and contribute to neurotoxicity and neurodegeneration APP also interacts with various proteins to mediate intracellular signaling and cellular responses Mutations in the APP gene are associated with early-onset familial Alzheimer’s disease, highlighting its role in the pathogenesis of neurodegenerative disorders
