Description
Dioxygenase that demethylates RNA by oxidative demethylation: specifically demethylates N(6)-methyladenosine(m6A) RNA, the most prevalent internal modification of messenger RNA(mRNA) in higher eukaryotes(PubMed:23177736, PubMed:24778178, PubMed:24616105, PubMed:24489119). Can also demethylate N(6)-methyladenosine in single-stranded DNA(in vitro)(PubMed:24616105). Requires molecular oxygen, alpha-ketoglutarate and iron(PubMed:21264265, PubMed:23177736, PubMed:24778178, PubMed:24616105, PubMed:24489119). Demethylation of m6A mRNA affects mRNA processing and export(PubMed:23177736). Required for spermatogenesis(By similarity).

Descripción
ALKBH5 is an α-ketoglutarate (α-KG)- and Fe(II)-dependent dioxygenase that specifically catalyzes the demethylation of N6-methyladenosine (m6A), a major modification on mRNA. ALKBH5 is a key regulator of RNA metabolism, influencing mRNA stability, splicing, translation, and export. By removing m6A marks, ALKBH5 modulates gene expression, particularly during processes such as spermatogenesis, immune responses, and cancer progression. ALKBH5 is highly expressed in testicular tissues, where it regulates germ cell development and fertility. In cancer biology, ALKBH5 plays a critical role in tumor progression and metastasis by promoting the stability and translation of oncogenes and cancer-associated transcripts. Its upregulation has been observed in glioblastoma, breast cancer, and other tumors, where it contributes to chemoresistance and proliferation. Recent studies highlight ALKBH5 as a potential therapeutic target for disrupting cancer-associated RNA modifications and inhibiting tumor growth.
