Description

Progesterone-dependent acylglycerol lipase that catalyzes hydrolysis of endocannabinoid arachidonoylglycerol(AG) from cell membrane(PubMed:26989199). Acts as a progesterone receptor: progesterone-binding activates the acylglycerol lipase activity, mediating degradation of 1-arachidonoylglycerol(1AG) and 2-arachidonoylglycerol(2AG) to glycerol and arachidonic acid(AA)(PubMed:26989199). Plays a key role in sperm capacitation in response to progesterone by mediating degradation of 2AG, an inhibitor of the sperm calcium channel CatSper, leading to calcium influx via CatSper and sperm activation(PubMed:26989199). May also play a role in smooth muscle cells migration(By similarity).

Descripción

ABHD2 is a serine hydrolase well-known for its role in sperm activation and fertilization by regulating the CatSper calcium channel, which is activated by progesterone. This calcium influx is essential for sperm motility and the acrosome reaction. Beyond reproduction, ABHD2 is expressed in the lungs, brain, and immune cells, where it hydrolyzes bioactive lipids, including 2-arachidonoylglycerol (2-AG), a key endocannabinoid. Through lipid metabolism, ABHD2 modulates inflammatory responses, immune signaling, and cellular lipid composition. Dysregulation of ABHD2 has been associated with infertility, due to impaired sperm activation, and inflammatory diseases, highlighting its dual importance in reproductive biology and immune regulation. Structurally, ABHD2 features a catalytic triad within its hydrolase domain, driving its activity in lipid processing and signaling.
