Descripción

AMOTL2 is a member of the angiomotin (AMOT) protein family which regulates cellular processes such as angiogenesis, cell polarity, migration, and proliferation. AMOTL2 interacts with components of the Hippo signaling pathway specifically binding to YAP (Yes-associated protein) and TAZ (transcriptional co-activator with PDZ-binding motif) to regulate gene expression and maintain tissue homeostasis. By inhibiting YAP/TAZ nuclear translocation AMOTL2 acts as a tumor suppressor controlling cell proliferation and limiting excessive tissue growth. AMOTL2 plays a critical role in vascular development by stabilizing endothelial cell junctions and maintaining endothelial barrier integrity during angiogenesis. It interacts with cytoskeletal components such as actin filaments to mediate endothelial cell polarity and migration processes that are essential for blood vessel formation. AMOTL2 also regulates epithelial cell polarity during embryonic development influencing organogenesis and tissue architecture. In cancer biology AMOTL2 has dual roles depending on the cellular context. It can function as a tumor suppressor by restricting oncogenic YAP/TAZ signaling and inhibiting cell migration while in some cancers its dysregulation may promote tumor progression, metastasis, and angiogenesis. AMOTL2’s ability to control cell-cell junction stability and its involvement in actin cytoskeleton remodeling make it a key regulator of cellular structure and migration.
