Descripción
ANAPC4 is a key structural subunit of the Anaphase-Promoting Complex/Cyclosome (APC/C), a multi-subunit E3 ubiquitin ligase that controls the progression of the cell cycle by targeting regulatory proteins for proteasomal degradation. ANAPC4 interacts with other core components, including ANAPC1 and ANAPC5, to form the scaffold that supports APC/C assembly and its enzymatic function. ANAPC4 plays an essential role in facilitating the ubiquitination of substrates such as cyclin B and securin, ensuring their degradation for the metaphase-to-anaphase transition and mitotic exit. Its proper function is necessary for maintaining chromosomal stability and preventing aneuploidy. Dysregulation of ANAPC4 leads to impaired APC/C activity, causing mitotic defects and genomic instability, which are strongly associated with tumorigenesis. ANAPC4 is emerging as a critical factor for cell division and a potential target for therapeutic intervention in proliferative diseases such as cancer.

A large protein complex, termed the anaphase-promoting complex (APC), or the cyclosome, promotes metaphase-anaphase transition by ubiquitinating its specific substrates such as mitotic cyclins and anaphase inhibitor, which are subsequently degraded by the 26S proteasome. Biochemical studies have shown that the vertebrate APC contains eight subunits. The composition of the APC is highly conserved in organisms from yeast to humans.
