Alpha- and Gamma-Adaptin-Binding Protein P34 (AAGAB) is a cytoplasmic protein that interacts with adaptin proteins, which are part of the clathrin-mediated endocytosis machinery. AAGAB plays a crucial role in regulating vesicular trafficking, including the internalization and recycling of cell surface receptors and membrane proteins. It is involved in maintaining cellular homeostasis by mediating the transport of molecules between the plasma membrane, endosomes, and other intracellular compartments. Mutations in the AAGAB gene are linked to palmoplantar keratoderma (PPK), a skin disorder characterized by hyperkeratosis of the palms and soles, due to defective vesicular transport and keratinocyte function. Emerging research suggests that AAGAB may also influence cell signaling and proliferation through its role in membrane protein trafficking. Its importance in cellular dynamics highlights its potential as a target for understanding and treating vesicle transport-related disorders.
