Description

ACER3 is an endoplasmic reticulum-localized ceramidase that specializes in hydrolyzing unsaturated ceramides into sphingosine and free fatty acids, optimizing its activity in alkaline pH environments. It is highly expressed in metabolically active and neuronal tissues, such as the brain, liver, and pancreas, where it regulates sphingolipid metabolism and supports processes such as cell survival, apoptosis, autophagy, and stress responses. ACER3 plays a particularly critical role in the nervous system, where sphingolipid homeostasis is essential for neuronal function and survival. Mutations in the ACER3 gene have been associated with hereditary spastic paraplegia, a rare neurodegenerative condition marked by spasticity and progressive weakness. Additionally, ACER3 is implicated in metabolic diseases and cancer, where its regulation of sphingolipids impacts key signaling pathways. By balancing ceramide and sphingosine levels, ACER3 supports cellular adaptation to stress and maintains bioactive lipid equilibrium, making it a potential therapeutic target for neurodegenerative disorders, metabolic dysfunctions, and tumor progression.
