Descripción
The ALK (anaplastic lymphoma kinase) gene encodes a receptor tyrosine kinase, which is a type of cell surface receptor that plays a role in signaling pathways involved in cell growth, proliferation, and differentiation. The ALK protein belongs to the insulin receptor superfamily and is normally expressed in specific tissues during embryonic development, where it helps regulate normal cell growth and development. While ALK expression is typically limited to certain tissues during development, aberrant activation of the ALK gene can occur in various cancers due to chromosomal rearrangements, gene amplification, or activating mutations. These alterations lead to the overexpression or constitutive activation of the ALK protein, contributing to oncogenesis (the formation of cancer).The most well-known oncogenic alteration involving the ALK gene is the fusion of the ALK gene with another gene, resulting in a chimeric fusion protein with constitutive kinase activity. This fusion protein can drive cellular transformation and tumorigenesis.

Anaplastic Lymphoma Kinase (ALK), also known as CD246, is a 220 kDa transmembrane glycoprotein that is primarily expressed in the developing nervous system. ALK contains one LDL Receptor (LDLR) class A domain, two meprin/A5-protein/PTPmu (MAM) domains, and a cytoplasmic kinase domain. Chromosomal translocation involving ALK and the nucleolar NPM protein result in a cytoplasmic fusion protein that exists as a dimer. The kinase domain in NPM-ALK is constitutively active and participates in cellular transformation. ALK translocations are found in a variety of tumors. Within the extracellular domain, human and mouse ALK share 88% amino acid sequence identity.

ALK is usually found in the nervous system and appears to play an important role in the normal development and function of the nervous system. ALK was originally identified as part of the NPM(Nucleophosmin)-ALK oncogenic fusion protein, resulting from the(2;5)(p23;q35) translocation that is frequently associated with anaplastic large-cell lymphoma(ALCL). The EML4(echinoderm microtubule-associated protein-like 4)-ALK fusion protein have been described in non-small-cell lung cancer(NSCLC), this transforming fusion kinase is a promising candidate for a therapeutic target as well as for a diagnostic molecular marker in NSCLC(PMID: 17625570).
