Activator of apoptosis harakiri (HRK) is a pro-apoptotic protein that belongs to the Bcl-2 family of proteins, which regulate cell death by controlling mitochondrial outer membrane permeabilization HRK is involved in initiating apoptosis in response to cellular stress, including DNA damage, oxidative stress, and growth factor deprivation HRK interacts with anti-apoptotic proteins like Bcl-2 and Bcl-xL, promoting the release of apoptotic factors such as cytochrome c from the mitochondria, which activates caspases and leads to programmed cell death HRK is particularly important in neurons and immune cells, where it helps eliminate damaged or unwanted cells during development and in response to injury Dysregulation of HRK has been linked to various diseases, including cancer and neurodegenerative disorders, where loss of HRK expression allows cells to evade apoptosis and proliferate uncontrollably In cancer, HRK’s ability to promote cell death makes it a potential therapeutic target for inducing apoptosis in tumor cells

Activator of apoptosis Hrk regulates apoptosis through interaction with death-repressor proteins Bcl-2 and Bcl-X (L). The HRK protein lacks significant homology to other BCL2 family members except for an 8-amino acid region that was similar to the BCL2 homology domain-3 (BH3) motif of BIK. HRK interacts with BCL2 and BCLXL via the BH3 domain, but not with the death-promoting BCL2-related proteins BAX, BAK, or BCLXS.
