Background

ABHD5, also known as CGI-58, is a critical coactivator of adipose triglyceride lipase (ATGL), a key enzyme in lipolysis. It promotes the hydrolysis of triglycerides into glycerol and free fatty acids, thereby regulating lipid storage and energy metabolism. ABHD5 is highly expressed in adipose tissue, liver, and muscles, where it plays an essential role in lipid mobilization and energy balance. Mutations in the ABHD5 gene are associated with Chanarin-Dorfman syndrome, a rare lipid storage disorder characterized by excessive lipid accumulation in various tissues. Beyond lipolysis, ABHD5 influences lipid signaling pathways and cellular lipid composition, affecting processes such as inflammation and metabolic homeostasis.
