Descripción

AKAP10, also known as dual-specific AKAP2 (D-AKAP2), is a dual-targeting scaffold protein that binds to PKA and other signaling proteins at cellular membranes, including mitochondria and plasma membranes. AKAP10 regulates PKA signaling in pathways related to cardiac function, mitochondrial biogenesis, and cellular energy metabolism. Polymorphisms in AKAP10, such as the I646V variant, are associated with altered heart rate, reduced heart rate variability, and an increased risk of arrhythmias, highlighting its importance in cardiovascular homeostasis. AKAP10 also interacts with mitochondrial proteins to regulate energy production, enhancing ATP synthesis and oxidative phosphorylation. AKAP10 has been implicated in apoptosis, where its role in PKA-dependent signaling ensures mitochondrial integrity during cellular stress. Additionally, AKAP10 participates in cell migration and polarity by organizing signaling complexes near the plasma membrane, influencing cytoskeletal reorganization and adhesion dynamics in cancer metastasis.

Differentially targeted protein that binds to type I and II regulatory subunits of protein kinase A and anchors them to the mitochondria or the plasma membrane. Although the physiological relevance between PKA and AKAPS with mitochondria is not fully understood, one idea is that BAD, a proapoptotic member, is phosphorylated and inactivated by mitochondria-anchored PKA. It cannot be excluded too that it may facilitate PKA as well as G protein signal transduction, by acting as an adapter for assembling multiprotein complexes. With its RGS domain, it could lead to the interaction to G-alpha proteins, providing a link between the signaling machinery and the downstream kinase(By similarity).
