Descripción
VAV2 is a ubiquitously expressed guanine nucleotide exchange factor (GEF) that activates Rho family GTPases, including Rac1, RhoA, and Cdc42, to regulate cytoskeletal dynamics, cell migration, and intracellular signaling pathways. Unlike VAV1, VAV2 functions in both hematopoietic and non-hematopoietic cells, where it plays roles in integrin signaling, epithelial cell polarity, and vascular development. VAV2 is activated downstream of receptor tyrosine kinases (RTKs) and integrins, linking extracellular cues to actin cytoskeleton remodeling and cell adhesion processes. Dysregulation of VAV2 is implicated in cancer progression, where it promotes cell invasion, metastasis, and epithelial-mesenchymal transition (EMT) by activating Rac1 signaling. Knockout studies reveal defects in vascular development, impaired epithelial integrity, and reduced cell migration, underscoring its role in cytoskeletal reorganization, integrin signaling, and tissue homeostasis.
