Descripción
TMED1, a member of the p24 protein family, is a type I transmembrane protein involved in vesicular trafficking, particularly in the early secretory pathway. TMED1 localizes to the endoplasmic reticulum (ER), ER-Golgi intermediate compartment (ERGIC), and the Golgi apparatus, where it regulates the transport and sorting of cargo proteins between cellular compartments. It interacts with coat protein complexes (COPI and COPII) to facilitate vesicle budding, cargo selection, and trafficking efficiency. TMED1 also plays a role in cellular signaling and homeostasis through its interaction with membrane-bound receptors and secretory proteins. It is widely expressed in various tissues, particularly in secretory cells and immune tissues, where it supports protein maturation, secretion, and immune regulation. Dysregulation of TMED1 has been linked to cancers and immune dysfunction, where altered trafficking pathways impair protein sorting and cellular communication. Knockdown studies show defects in vesicle formation, impaired protein transport, and disrupted Golgi integrity, emphasizing its role in maintaining secretory pathway dynamics, intracellular signaling, and cellular homeostasis.
