TNFRSF1A Associated Via Death Domain (TRADD) is an adaptor protein critical for tumor necrosis factor (TNF)-mediated signaling. It acts as a central platform for assembling signaling complexes upon TNF receptor 1 (TNFR1) activation. TRADD mediates divergent pathways leading to apoptosis, necroptosis, or NF-κB activation, depending on the recruited proteins. It plays a pivotal role in immune responses, inflammation, and cell death regulation. Aberrations in TRADD function are linked to immune dysregulation, chronic inflammation, and cancer. Its dual role in promoting cell survival and death makes it a crucial target for therapeutic strategies.

The nuclear form acts as a tumor suppressor by preventing ubiquitination and degradation of isoform p19ARF/ARF of CDKN2A by TRIP12: acts by interacting with TRIP12, leading to disrupt interaction between TRIP12 and isoform p19ARF/ARF of CDKN2A(By similarity).
