Descripción

The Thyrotropin-Releasing Hormone Receptor (TRHR) is a G protein-coupled receptor (GPCR) that is activated by thyrotropin-releasing hormone (TRH), a peptide hormone responsible for regulating the secretion of thyroid-stimulating hormone (TSH) from the pituitary gland. TRHR is primarily expressed in the anterior pituitary, where it plays a critical role in the regulation of thyroid function by stimulating the release of TSH. Upon activation, TRHR couples to Gq proteins, stimulating phospholipase C (PLC) and increasing intracellular calcium levels, which in turn promotes the release of TSH and the synthesis of thyroid hormones (T3 and T4). TRHR also plays a role in regulating metabolic processes, growth, and development. Dysregulation of TRHR signaling is implicated in disorders such as hypothyroidism, hyperthyroidism, and thyroid tumors. Targeting TRHR with agonists or antagonists can be useful for managing thyroid-related conditions and regulating thyroid hormone production.

Thyrotropin-releasing hormone (TRH; MIM 275120), a small neuropeptide, is widely distributed throughout the central and peripheral nervous system as well as in extraneural tissues. The peptide is synthesized in the hypothalamus and transported by the portal vascular system to the anterior pituitary where it acts on thyrotropic and lactotropic cells to promote secretion of TSH and prolactin, respectively. Thyrotropin-releasing hormone receptor is a G protein-coupled receptor that activates the inositol phospholipid-calcium-protein kinase C transduction pathway upon the binding of TRH. The TRHR gene is expressed in the thyrotrope cells of the anterior pituitary.
