Descripción

The thromboxane A2 receptor (TBXA2R), also known as TP, is a G protein-coupled receptor activated by thromboxane A2 (TXA2), a lipid mediator that promotes vasoconstriction, platelet aggregation, and inflammation. TBXA2R primarily couples to Gq and G12/13 proteins, activating phospholipase C (PLC), increasing intracellular calcium levels, and triggering Rho-mediated pathways that regulate smooth muscle contraction and platelet activation. TBXA2R is highly expressed in platelets, vascular smooth muscle cells, and immune cells, where it plays a pivotal role in hemostasis, thrombosis, and vascular tone. Dysregulation of TBXA2R signaling contributes to thrombotic disorders, hypertension, and atherosclerosis. TBXA2R antagonists, such as terutroban, are being developed as antithrombotic agents to reduce platelet aggregation and treat cardiovascular diseases. Targeting TBXA2R provides therapeutic potential for managing thrombosis, pulmonary hypertension, and inflammatory vascular diseases by modulating TXA2-mediated effects.
