S1PR5 is a G protein-coupled receptor for sphingosine-1-phosphate (S1P), predominantly expressed in oligodendrocytes, natural killer (NK) cells, and endothelial cells. S1PR5 couples to Gi and G12/13 proteins, regulating pathways involved in cell migration, proliferation, and survival. In the central nervous system, S1PR5 plays a critical role in oligodendrocyte differentiation and myelination, contributing to neuronal protection and repair. It also regulates NK cell trafficking and activity, enhancing immune surveillance and anti-tumor responses. Dysregulation of S1PR5 signaling is implicated in neurodegenerative diseases, such as multiple sclerosis, where impaired myelination contributes to disease pathology. In cancer, S1PR5 expression is linked to immune evasion and tumor progression, particularly through modulation of NK cell function. S1PR5 agonists are being developed as therapies for multiple sclerosis to promote remyelination and reduce neuroinflammation, while its role in immune cell regulation makes it a promising target for cancer immunotherapy and inflammatory diseases.
