AK5 is a cytosolic adenylate kinase predominantly expressed in the brain, where it plays a role in neural energy metabolism and synaptic function. Unlike other AK isoforms, AK5 demonstrates substrate specificity for ATP/GTP and AMP. It is thought to regulate energy transfer and storage in neurons, particularly in response to neurotransmission demands. AK5 expression correlates with cognitive functions, and its dysfunction has been implicated in neurodevelopmental disorders and memory deficits. AK5 has been shown to influence energy-sensing pathways, such as AMPK, thereby contributing to neuronal survival during energy depletion.
