Background
Nuclear receptor subfamily 1 group D member 2 (NR1D2), also known as Rev-Erbβ, is a closely related homolog of NR1D1, and it also plays a crucial role in regulating circadian rhythms and metabolic homeostasis NR1D2 is involved in controlling the expression of genes that regulate the body’s internal clock, metabolism, and immune responses Similar to NR1D1, NR1D2 acts by repressing the transcription of target genes through binding to specific DNA elements, influencing lipid and glucose metabolism, and modulating inflammatory responses NR1D2 is highly expressed in tissues such as the liver, skeletal muscle, and adipose tissue, where it helps control circadian variations in metabolism and energy expenditure Dysregulation of NR1D2 has been associated with metabolic disorders, including obesity, type 2 diabetes, and fatty liver disease, as well as inflammation-related conditions like autoimmune diseases and cardiovascular disorders In addition to its role in metabolism and immune function, NR1D2’s participation in regulating the circadian rhythm highlights its potential therapeutic value for managing disorders related to disrupted biological clocks, such as sleep disturbances and shift work-induced metabolic abnormalities
