Background

Amyloid beta (Aβ or Abeta) is a term used to describe peptides composed of 36 to 43 amino acids, and they constitute the primary component of the amyloid plaques found within the brains of individuals suffering from Alzheimer's disease. These peptides are derived from the amyloid-beta precursor protein (APP).The formation of β-amyloid occurs through the sequential cleavage of a transmembrane protein known as APP, which is a glycoprotein serving as its precursor. Among the various forms, the most extensively studied variants are Aβ42 and Aβ40 (Abeta40 and Abeta42). The sole distinction between Aβ42 and Aβ40 is the presence of two additional residues at the C-terminus of Aβ42.The generation of Aβ from its precursor protein, APP, is orchestrated by enzymes known as gamma- and β-secretases. Aβ can be found circulating in plasma, cerebrospinal fluid (CSF), and brain interstitial fluid (ISF), predominantly in the soluble Aβ40 form. Notably, amyloid plaques contain both Aβ40 and Aβ42.The amyloid-beta peptide is believed to play a central role in the pathology of Alzheimer's disease. Sporadic Alzheimer's disease is the most prevalent cause of dementia, contributing to an estimated 50% to 56% of cases in autopsy and clinical series.Furthermore, Aβ production appears to have a close relationship with the Aβ precursor protein, APP, and mitochondria, which serve as the primary energy source for the brain. Microglial cells also play crucial roles in removing accumulated Aβ, not only through phagocytosis but also by releasing proteases for degradation. Beta amiloide (Aβ o Abeta) es un término utilizado para describir péptidos compuestos de 36 a 43 aminoácidos y constituyen el componente principal de las placas amiloides que se encuentran en el cerebro de personas que padecen la enfermedad de Alzheimer. Estos péptidos se derivan de la proteína precursora del beta-amiloide (APP). La formación del β-amiloide se produce mediante la escisión secuencial de una proteína transmembrana conocida como APP, que es una glicoproteína que actúa como su precursora. Entre las diversas formas, las variantes más estudiadas son Aβ42 y Aβ40 (Abeta40 y Abeta42). La única distinción entre Aβ42 y Aβ40 es la presencia de dos residuos adicionales en el extremo C de Aβ42. La generación de Aβ a partir de su proteína precursora, APP, está orquestada por enzimas conocidas como gamma y β-secretasas.
