RAB35, member RAS oncogene family (RAB35) is a small GTPase involved in regulating endocytosis, vesicle trafficking, and membrane recycling in the cell It plays an essential role in the trafficking of cellular membranes and proteins, particularly in the sorting of endosomal cargo for recycling or degradation RAB35 is also crucial in the regulation of autophagy, where it helps in the formation of autophagosomes for the degradation of dysfunctional cellular components This GTPase is involved in cellular signaling by modulating the movement of receptors and other signaling molecules to and from the cell membrane, thus influencing cell communication, adhesion, and migration RAB35’s dysfunction can result in impaired cellular homeostasis, contributing to diseases such as cancer, where disrupted vesicular trafficking supports uncontrolled cell growth and metastasis Moreover, defects in RAB35 are also linked to neurodegenerative conditions, where defective vesicular trafficking leads to cellular stress and dysfunction

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different sets of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion. That Rab is involved in the process of endocytosis and is an essential rate-limiting regulator of the fast recycling pathway back to the plasma membrane.
