Biological Activity: A microchemotaxis assay was a carried out to determine the chemotactic effect of IL8 on Jurkat cells. 100 µm cell suspension (1 × 106 cells per ml) in RPMI 1640 FBS free medium were seeded to the upper chamber of a microchemotaxis system and Human IL8 (0.1 ng/ml, 1 ng/ml and 10 ng/ml dilutions in RPMI 1640) was added to the lower chamber with an 8 µm polycarbonate filter to separate the two compartments. The cells were incubated at 5% CO2, 37°C for 1 hour, the filter was removed and the cells in the lower chamber were observed at x100 magnification and counted at x400 magnification. It was determined that optimal chemotaxis of Jurkat cells occurs at 1-10 ng/ml concentrations of Human IL8.

The stock concentration should be between 0.1-1.0 mg/ml. Do not vortex.
