Descripción

PHF19 is a chromatin-associated protein that contains a PHD finger domain and functions as a component of the Polycomb repressive complex 2 (PRC2). It regulates gene expression through the methylation of histone H3 on lysine 27 (H3K27me3), contributing to transcriptional repression and chromatin compaction. PHF19 acts as a recruiter of PRC2 to actively transcribed genes, linking gene activation to repression during processes like cellular differentiation, proliferation, and stem cell maintenance. It is highly expressed in embryonic stem cells and proliferative tissues, where it plays roles in developmental programming and epigenetic control. Dysregulation of PHF19 has been linked to cancers, including lymphoma and glioblastoma, where it promotes tumorigenesis through aberrant PRC2 activity and disrupted gene silencing. Knockdown studies reveal impaired PRC2 recruitment, altered histone methylation, and disrupted differentiation, highlighting its role in chromatin regulation, transcriptional repression, and developmental pathways.
