Descripción
TRAF6 is a multifunctional adaptor protein that regulates signaling downstream of TNFR superfamily members, Toll-like receptors (TLRs), and IL-1 receptors. It activates key pathways, including NF-κB, JNK, and Akt, to mediate inflammatory responses, innate immunity, and bone homeostasis. TRAF6 also functions as an E3 ubiquitin ligase, facilitating K63-linked ubiquitination of signaling intermediates like TAK1, IKK, and Akt to promote downstream activation. It plays essential roles in immune cell activation, osteoclast differentiation, and autophagy regulation. TRAF6 is widely expressed in immune cells, bone tissue, and epithelial cells, where it supports pathogen recognition, inflammatory signaling, and bone remodeling. Dysregulation of TRAF6 is associated with autoimmune diseases, chronic inflammation, osteoporosis, and cancer due to hyperactive NF-κB signaling and impaired bone resorption. Knockout models exhibit immune deficiencies, defective TLR signaling, impaired bone development, and increased susceptibility to infections, underscoring its critical role in immunity, inflammation, and skeletal homeostasis.
