Background

MTOR associated protein, LST8 homolog (MLST8), also known as mammalian lethal with SEC13 protein 8, is a core component of the mechanistic target of rapamycin (mTOR) complexes, mTORC1 and mTORC2. MLST8 is characterized by its seven WD40 repeats, forming a β-propeller structure that mediates interactions with other mTOR complex components. Within mTORC1, MLST8 contributes to substrate binding and mTOR kinase stability, enhancing its role in regulating cellular growth, metabolism, and response to nutrient availability. In mTORC2, MLST8 is crucial for the assembly and stabilization of the complex, which regulates cytoskeletal dynamics, cell survival, and metabolism. Dysregulation of MLST8 or its associated mTOR pathways has been linked to various diseases, including cancer, neurodegenerative disorders, and metabolic syndromes. MLST8's functional importance makes it a potential therapeutic target for modulating mTOR activity in disease contexts.
