Descripción
DENN domain containing 6B (DENND6B) is a closely related paralog of DENND6A and functions as a guanine nucleotide exchange factor (GEF) for Rab GTPases, particularly Rab14. DENND6B is critical in regulating vesicle trafficking and endosomal recycling by facilitating Rab14 activation, which supports the transport of cargo between endosomes and the Golgi network and promotes receptor recycling at the plasma membrane. This activity is essential for maintaining cellular signaling, protein trafficking, and epithelial cell polarity. DENND6B is expressed in a variety of tissues, with notable roles in epithelial cells and immune tissues, where vesicular transport is critical for cellular communication and immune response. Dysregulation of DENND6B has been implicated in pathological conditions such as cancer metastasis and immune dysfunction, where defects in vesicular trafficking disrupt cellular adhesion, signaling, and migration. Its functional similarity to DENND6A emphasizes the cooperative roles of these proteins in intracellular transport and maintaining cellular integrity.
