Background

JAM3 is predominantly expressed in endothelial cells, platelets, and certain subsets of immune cells. Like JAM2, it features two extracellular Ig-like domains, a single transmembrane domain, and a cytoplasmic tail that interacts with scaffolding proteins to organize tight junctions. JAM3 plays a critical role in endothelial cell adhesion and platelet aggregation, as well as in the regulation of vascular permeability and angiogenesis. It interacts with other adhesion molecules, such as JAM2 and integrins, to mediate cell-cell interactions and leukocyte trafficking during immune responses. Additionally, JAM3 is essential for maintaining the integrity of the blood-brain barrier and regulating cell migration in response to injury or inflammation.
