The interleukin-9 receptor (IL9R) is a key component of the immune system, primarily involved in mediating the biological activities of interleukin-9 (IL-9), a cytokine secreted by T helper 2 (Th2) cells, mast cells, and a subset of T helper cells known as Th9 cells. IL-9 is known for its pleiotropic effects, influencing various cell types including mast cells, T cells, B cells, and epithelial cells. It is particularly important in immune responses, inflammatory processes, and allergic reactions. IL9R, through binding IL-9, initiates signaling cascades that play critical roles in the regulation of cell survival, proliferation, and differentiation in immune responses. The IL9R gene is located on the X chromosome (Xq28) in humans, which makes certain IL9R-related mutations have potential X-linked patterns of inheritance. Notably, IL9R signaling is involved in allergic diseases, asthma, and certain cancers. Additionally, IL-9 and IL9R have been implicated in autoimmune diseases and responses to infections, such as helminth infections. Due to its versatile role in immunity, IL9R is an area of interest for therapeutic strategies aiming to modulate immune responses, particularly in allergy and asthma, and cancer treatment.

The protein encoded by this gene is a cytokine receptor that specifically mediates the biological effects of interleukin 9 (IL9). The functional IL9 receptor complex requires this protein as well as the interleukin 2 receptor, gamma (IL2RG), a common gamma subunit shared by the receptors of many different cytokines. The ligand binding of this receptor leads to the activation of various JAK kinases and STAT proteins, which connect to different biologic responses. This gene is located at the pseudoautosomal regions of X and Y chromosomes. Genetic studies suggested an association of this gene with the development of asthma. Multiple pseudogenes on chromosome 9, 10, 16, and 18 have been described. Alternatively spliced transcript variants have been found for this gene.
