Estrogen receptor 1 (ESR1), also known as ERα, is a nuclear receptor that mediates the effects of estrogen, a key hormone in regulating the female reproductive system and other physiological processes ESR1 is expressed in a variety of tissues, including the breast, uterus, brain, and bones, where it regulates gene expression related to reproductive health, bone metabolism, and brain function Upon binding to estrogen, ESR1 undergoes a conformational change, allowing it to bind to estrogen response elements (EREs) on target genes and modulate their expression This activation regulates processes such as cell proliferation, differentiation, and apoptosis, and it is essential for the normal development of the reproductive organs and the maintenance of bone density Dysregulation of ESR1 has been linked to various diseases, particularly hormone receptor-positive breast cancer, where overactivation of ESR1 signaling promotes tumor cell growth and survival ESR1 is also a therapeutic target in breast cancer, with drugs like selective estrogen receptor modulators (SERMs) used to block or modulate its activity in cancer treatment
